Two years after the first film, Blade learns that Whistler didn't die: Frost turned him into a vampire and he's been held prisoner. Blade finds him in a tank in Prague and cures him with a blood treatment. Whistler comes back a little suspicious, and things get tense with Scud, the young tech guy who now helps Blade with weapons.
The vampire world is facing a new threat: the Reapers. They're mutants born from a virus. Their jaws split open in three parts, their hunger never ends and they bite humans and vampires alike, turning everyone into Reapers. The first one is Jared Nomak. Vampire overlord Eli Damaskinos offers a truce: Blade will lead the Bloodpack, an elite vampire squad that was trained to kill him. The team includes Damaskinos's daughter Nyssa and the hulking Reinhardt.
The hunt starts at a vampire nightclub and moves into the sewers of Prague. They find out the Reapers are immune to almost everything, garlic and silver included. Only sunlight works, and their hearts are shielded by bone that can only be pierced from the side. Several Bloodpack members die or get infected in the nest. Nomak finds Blade and offers an alliance against the vampires, but Blade turns him down.
Then comes the twist. Damaskinos created the Reapers on purpose, trying to build a vampire that could survive the sun. Nomak is his son, an experiment gone wrong. Scud turns out to be a spy working for the vampires, and Blade blows him up. Reinhardt betrays the team and Blade is captured so Damaskinos can study his blood. Nyssa, who has grown close to Blade, learns the truth and is torn.
Whistler frees Blade, who recovers by plunging into a pool of blood. He slices Reinhardt in half and fights his way to the lab. Nomak gets there first and kills his father. Nyssa, bitten by her brother, is doomed. Blade fights Nomak until the Reaper drives Blade's sword into the weak spot in his own chest and dies. Nyssa asks to see the sunrise while she's still a vampire, and Blade carries her outside, where she turns to ash in his arms. In the epilogue, in London, Blade settles the score with one of Damaskinos's men who got away.
